NM28-2746 is a multispecific, trispecific T‑cell engager antibody candidate developed using Numab’s MATCH4 platform that targets mesothelin (MSLN), CD3 on T cells, and human serum albumin (HSA) for half‑life extension.[1][2][3][4] It consists of four stabilized and humanized antibody variable domain fragments arranged in a heterodimeric, bivalent αMSLN configuration that confers avidity-based binding, enabling selective activation and redirection of T cells to kill high MSLN‑expressing tumor cells while sparing low‑MSLN normal tissues and retaining activity in the presence of high levels of soluble mesothelin.[1][2][3][4] The drug is being developed primarily for solid tumors that overexpress mesothelin, including mesothelioma and cancers of the lung, biliary tract, ovary, breast, and pancreas, with regional development and commercialization rights in Greater China licensed to 3SBio’s subsidiary Sunshine Guojian Pharmaceutical.[1][2]
