NM32-2668 is a trispecific antibody developed by Numab Therapeutics for the treatment of solid tumors and hematological malignancies. It targets three antigens: receptor tyrosine kinase-like orphan receptor 1 (ROR1), T-cell surface antigen CD3, and human serum albumin (HSA). The anti-ROR1 domain directs the antibody to tumor cells overexpressing ROR1, which is associated with poor prognosis in cancers such as chronic lymphocytic leukemia, mantle cell lymphoma, triple-negative breast cancer, lung cancer, and ovarian cancer. The anti-CD3 domain engages T cells to mediate targeted tumor cell lysis. Binding to HSA extends the half-life of the molecule in circulation for more convenient dosing. Preclinical studies show that NM32-2668 induces specific T cell-mediated killing of both dividing and non-dividing ROR1-positive tumor cells while minimizing cytokine release compared to other CD3 engagers. The drug is currently in Phase 1 clinical trials for various solid tumors[1][2][3][5][6][7].
