Overview

NM5072 is an investigational monoclonal antibody developed by NovelMed Therapeutics for the treatment of paroxysmal nocturnal hemoglobinuria (PNH), a rare, chronic, inflammatory, and hemolytic disease characterized by the destruction of red blood cells leading to anemia and other severe symptoms. The drug acts as an anti-properdin antibody that selectively inhibits properdin—a key protein in the alternative complement pathway—while preserving the classical complement pathway necessary for immune defense against infections. This selective inhibition prevents lysis of PNH red blood cells and aims to improve anemia without compromising infection protection. NM5072 has completed a Phase 1 trial in healthy volunteers demonstrating safety and dose-dependent inhibition of the alternative pathway, with plans for Phase 2 trials in PNH patients as well as other complement-mediated diseases such as C3 glomerulopathy and atypical hemolytic uremic syndrome[1][2][3][4][5][7].
