Overview

NMA3 is a rat-derived monoclonal antibody clone targeting the extracellular domain of nicastrin (NCT), a key structural and functional component of the gamma-secretase enzyme complex. Developed by researchers at Imperial College London, NMA3 was generated via genetic immunization with nicastrin cDNA. Nicastrin plays a critical role in gamma-secretase substrate recognition, which in turn activates oncogenic signaling pathways such as Notch, CD44, and EpCAM. While other clones from the same program (such as NMA2 and NMA4) demonstrated potent functional inhibition of breast cancer cell invasion and proliferation in vitro, NMA3 exhibited lower binding affinity to endogenous cell-surface nicastrin and was less functionally active in preclinical models of invasive breast cancer.
