NMBS-2 is a small-molecule inhibitor of hematopoietic progenitor kinase 1 (HPK1), also known as mitogen-activated protein kinase kinase kinase kinase 1 (MAP4K1). Developed by Nimbus Therapeutics, it is designed to function as a cancer immunotherapy by blocking HPK1, a negative regulator of T-cell receptor (TCR) signaling. By inhibiting HPK1, NMBS-2 enhances T-cell activation, restores cytokine production in exhausted T cells, and promotes anti-tumor immune responses. Preclinical data demonstrated that NMBS-2 effectively inhibits tumor growth both as a single agent and in combination with anti-PD-1 antibodies in various syngeneic mouse models.
