Drug intelligence / Profile preview
NMC-521 is a humanized monoclonal antibody developed by NomoCan Pharmaceuticals that selectively targets NMC-1, a novel cancer-specific antigen representing tumor-restricted splice variants of the MDM2 (HDM2) oncogene. Unlike normal cells, aggressive cancer cells aberrantly express these MDM2 variants on their plasma membrane. NMC-521 binds to these membrane-bound targets, inducing tumor cell death and natural killer (NK) cell-mediated cytotoxicity, which is accompanied by elevated levels of IL-15 and NK cell-associated RNAs. NMC-521 is in preclinical development for the treatment of aggressive solid tumors, including colorectal cancer, pancreatic cancer, and ocular cancers. NomoCan is also developing an antibody-drug conjugate (ADC) version of this candidate.
