NMD-01 is a first-in-class, investigational small molecule inhibitor of the skeletal muscle-specific chloride ion channel ClC-1. Developed by NMD Pharma, it is designed to enhance neuromuscular transmission and restore skeletal muscle function in patients with neuromuscular diseases. By inhibiting ClC-1, NMD-01 amplifies the muscle’s responsiveness to weak signals, thereby improving muscle excitability and function. This mechanism has shown clinical evidence of benefit in myasthenia gravis and preclinical efficacy in spinal muscular atrophy (SMA), Charcot-Marie-Tooth disease (CMT), and sarcopenia. The drug is administered orally and has demonstrated safety and pharmacological activity in Phase 1 trials with healthy volunteers[2][4][6][7][8].
