NMPR-047 is a synthetic protein-drug conjugate (PDC) being developed by New Path Molecular Research for the treatment of solid tumors, specifically colorectal adenocarcinoma and pancreatic ductal adenocarcinoma. The drug targets phosphatidylserine (PS), an anionic phospholipid that is typically sequestered on the inner leaflet of the plasma membrane in healthy cells but becomes exposed on the outer surface of tumor cells and tumor-associated vascular endothelial cells, serving as a marker of the tumor microenvironment and an immunosuppressive signal. NMPR-047 utilizes an SN-38 payload, the active metabolite of irinotecan, to induce DNA damage and cell death in targeted tissues. It is currently in the preclinical stage, with a target for IND/CTA readiness in the first half of 2028.
