Drug intelligence / Profile preview
NMRA-1 is a novel, orally bioavailable, non-selective positive allosteric modulator (PAM) of the N-methyl-D-aspartate receptor (NMDAR) that acts equipotently across the GluN2A, GluN2B, GluN2C, and GluN2D subunits. Developed by Neumora Therapeutics, NMRA-1 is designed to enhance physiological NMDAR function and restore dysregulated glutamatergic signaling, which is a key hypothesis underlying the etiology of schizophrenia. In preclinical studies, NMRA-1 demonstrated robust central nervous system activity, including the reduction of cortical gamma power in pharmaco-EEG models and the reversal of phencyclidine (PCP)-induced recognition memory deficits in rats, while maintaining a favorable tolerability profile with no significant effects on locomotor activity.
