Overview

NMRA-323511 is an investigational small molecule drug developed by Neumora Therapeutics for the treatment of agitation associated with dementia due to Alzheimer's disease. It acts as a highly potent and selective antagonist of the vasopressin 1a receptor (V1aR), exhibiting over 3,000-fold selectivity against V1b and V2 receptors and approximately 300-fold selectivity over the oxytocin receptor in preclinical studies. The V1aR is implicated in regulating aggression, affiliation, stress response, and anxiety; antagonism at this receptor is believed to have therapeutic potential for reducing symptoms of agitation in neuropsychiatric disorders. As of mid-2025, NMRA-323511 is being evaluated in Phase 1 clinical trials involving healthy elderly participants and adults with agitation related to Alzheimer's disease[2][3][4][5][6][7].
