NMS-E973 is a potent, selective, and orally available small molecule inhibitor of Heat Shock Protein 90 (Hsp90), a molecular chaperone essential for the stability and maturation of numerous oncogenic client proteins. Developed by Nerviano Medical Sciences, NMS-E973 has demonstrated nanomolar potency in preclinical models of various cancers, including glioblastoma and solid tumors. By inhibiting Hsp90, the drug induces the degradation of client proteins involved in cell signaling, proliferation, and survival, thereby exerting anti-tumor effects. It is currently being evaluated in preclinical studies, often in combination with standard-of-care treatments like temozolomide and radiotherapy, to explore potential synergistic therapeutic benefits.
