NMS-P114 (formerly RXDX-107) is a preclinical antibody-drug conjugate (ADC) developed by Nerviano Medical Sciences (NMS) targeting Nectin-4 (Poliovirus receptor-related protein 4), a cell surface adhesion molecule highly expressed in various solid tumors. The ADC utilizes NMS's proprietary site-specific conjugation technology to link a Nectin-4-targeting monoclonal antibody to a "next-generation anthracycline" payload, specifically a derivative of the potent DNA intercalator PNU-159682. Upon binding to Nectin-4 and subsequent internalization, the anthracycline payload is released, where it acts by intercalating into DNA and inhibiting topoisomerase II, leading to double-strand breaks and apoptosis. NMS-P114 has demonstrated significant antitumor activity in multiple preclinical models of Nectin-4-positive malignancies, including urothelial, triple-negative breast (TNBC), lung, gastric, esophageal, prostate, and head and neck cancers.
