NMS-P285 is a potent and highly selective small molecule inhibitor of BRAF kinase, belonging to the arylthiazole chemical class. Developed by Nerviano Medical Sciences, it acts as a Type I 1/2 inhibitor by occupying the ATP-binding pocket of the activated kinase (DFG-in conformation) and partly filling the kinase back pocket. NMS-P285 demonstrates strong antiproliferative activity against cancer cell lines harboring BRAF V600E or V600D mutations by blocking the MAPK signaling pathway (RAF-MEK-ERK). In preclinical models, it has shown superior antitumor activity and tumor regression compared to first-generation BRAF inhibitors like vemurafenib and dabrafenib, with a favorable pharmacokinetic profile following oral administration. As of 2012, it was identified as a candidate suitable for preclinical development.
