Overview

NMTi-ADC targeting HER2 is an investigational antibody-drug conjugate (ADC) developed by Myricx Bio. This drug consists of a selective and ultrapotent N-myristoyltransferase inhibitor (NMTi) payload conjugated via a cleavable linker to trastuzumab, a monoclonal antibody that targets the human epidermal growth factor receptor 2 (HER2). The mechanism of action involves targeted delivery of the NMT inhibitor to HER2-expressing tumor cells, leading to inhibition of protein myristoylation—a process essential for cancer cell survival and proliferation. Preclinical studies have demonstrated complete efficacy and encouraging tolerability in rodent and non-human primate models. The primary indication under investigation is HER2-positive cancers[1][4].
