Overview

NN-101 is a humanized single-domain camelid antibody (nanobody) developed by NanoNewron as part of its NewroBus platform technology. It is specifically engineered to target the transferrin receptor 1 (TfR1) expressed on brain endothelial cells, facilitating the transport of therapeutic biologics across the blood-brain barrier (BBB) through receptor-mediated transcytosis. Unlike some other TfR1-targeting agents, NN-101 is designed to bind to a site that does not compete with endogenous transferrin, thereby reducing the risk of hematological toxicity and interference with iron metabolism. Licensed from Rutgers University, this platform is intended to serve as a modular shuttle for delivering various payloads, such as enzymes or other antibodies, into the central nervous system to treat a range of neurodegenerative and CNS diseases. It is currently in the preclinical stage of development.
