NN-224 is a humanized TNF-α inhibitory nanobody (single-domain antibody) being developed by NanoNewron for the potential treatment of Alzheimer's disease and other neurodegenerative conditions. As a biologic therapeutic, it targets tumor necrosis factor-alpha (TNF-α), a pro-inflammatory cytokine that plays a central role in neuroinflammation and the progression of neurodegenerative diseases. NN-224 has demonstrated potent TNF-α inhibitory activity with an IC50 of 64.31 pM and exhibits high humanness and solubility scores. It is currently in preclinical development, positioned as a lead candidate alongside NN-223, and is designed to be compatible with NanoNewron's blood-brain barrier (BBB) crossing technology (NewroBus) for effective CNS delivery.
