NN-841 is a bi-functional humanized nanobody therapeutic developed to treat neuroinflammatory and neurodegenerative diseases, with a primary focus on Alzheimer’s disease. The molecule is a trimer composed of two nanobodies targeting tumor necrosis factor alpha (TNF-alpha) and one NewroBus™ nanobody that binds transferrin receptor 1 (TfR1). This design enables efficient transcytosis across the blood-brain barrier, allowing robust TNF-alpha inhibition within the central nervous system—a capability not achieved by current biologic TNF-alpha inhibitors. Preclinical studies have shown high inhibition of TNF-alpha activity in the brain and excellent tolerability when administered subcutaneously. NN-841 is currently advancing through preclinical evaluation, with IND-enabling studies planned. Its mechanism positions it as a potential treatment for Alzheimer’s disease and other CNS conditions characterized by elevated TNF-alpha levels, such as traumatic brain injury, multiple sclerosis, Huntington’s disease, Parkinson’s disease, epilepsy, and certain mood disorders[6][7][9].
