NN-843 is a bispecific nanobody heterotrimer developed by NanoNewron for the treatment of Alzheimer's disease. The drug candidate utilizes the proprietary NewroBus platform, which consists of nanobodies targeting the Transferrin Receptor 1 (TfR1) to facilitate transport across the blood-brain barrier (BBB) via receptor-mediated transcytosis. The therapeutic component of the construct is a nanobody designed to neutralize Tumor Necrosis Factor alpha (TNF-α), a pro-inflammatory cytokine that plays a central role in neuroinflammation and the progression of neurodegenerative diseases. By combining the BBB-crossing shuttle with a potent anti-TNF-α inhibitor, NN-843 aims to deliver therapeutic activity directly into the central nervous system to mitigate the neuroinflammatory processes associated with Alzheimer's disease. It is currently in the preclinical stage of development.
