Overview

NN3201 is a fully human monoclonal antibody-drug conjugate (ADC) that targets c-Kit (CD117, stem cell factor receptor), a receptor tyrosine kinase frequently overexpressed or mutated in various cancers. The ADC consists of the 2G4 antibody with high affinity for c-Kit, conjugated to the cytotoxic payload monomethyl auristatin E (MMAE) via a site-specific linker. Upon binding to c-Kit-expressing tumor cells, NN3201 is internalized and delivers MMAE intracellularly, resulting in tubulin inhibition and cell death. Preclinical studies demonstrate potent anti-tumor activity against both wild-type and mutant c-Kit positive tumors such as small cell lung cancer (SCLC) and gastrointestinal stromal tumors (GIST). Additional mechanisms may include modulation of the tumor microenvironment to enhance immune response. Developed by Novelty Nobility, Inc., NN3201 is currently being evaluated in Phase 1 clinical trials for advanced/metastatic solid tumors expressing c-Kit—including GIST, adenoid cystic carcinoma (ACC), uveal melanoma, neuroendocrine tumors (NET), chromophobe renal cell carcinoma (ChRCC), clear cell renal cell carcinoma (ccRCC), small cell lung cancer, and others[2][4][5][6][7][8].
