Overview

NNC 25-0926 is a research-stage small molecule glucagon receptor antagonist developed by Novo Nordisk for the treatment of type 2 diabetes. It functions by blocking the glucagon receptor, thereby inhibiting glucagon-stimulated hepatic glucose production (HGP) and reducing fasting hyperglycemia. Preclinical studies in conscious dogs and high-fat diet-fed mice have demonstrated that NNC 25-0926 dose-dependently inhibits glucagon-stimulated net hepatic glucose output and improves glucose tolerance. Chronic treatment has also been shown to improve islet sensitivity to glucose and augment insulin secretion, addressing key defects in type 2 diabetes.
