Overview

25-2648 (also known as NNC-25-2648) is a small molecule glucagon receptor antagonist (GRA) developed by Agouron Pharmaceuticals and Novo Nordisk. It selectively binds to and blocks the glucagon receptor (GCGR), inhibiting glucagon-mediated hepatic glucose production and regulating amino acid metabolism. Although initially developed as a therapeutic candidate for the treatment of type 2 diabetes, its clinical development was terminated. It remains widely utilized in preclinical research as a pharmacological tool to investigate the liver-alpha-cell axis, glucagon signaling, and metabolic pathways related to amino acid and lipid metabolism.
