NNC0114-0000-0005 is a fully human recombinant monoclonal immunoglobulin G1 (IgG1) antibody that binds to and neutralizes interleukin 21 (IL‑21). By targeting IL‑21, a cytokine implicated in the pathogenesis of autoimmune diseases such as rheumatoid arthritis (RA), this drug aims to modulate immune responses. It was developed for the treatment of several immune-mediated conditions including RA, Crohn's disease, systemic lupus erythematosus (SLE), and type 1 diabetes mellitus. The drug demonstrated linear pharmacokinetics with a mean terminal half-life of approximately two to three weeks and showed preliminary signs of reduced RA activity at higher doses in early clinical trials[1][3][4]. Development has been discontinued for all major indications.
