NNC0215-0384 is a pegylated, humanized monoclonal antibody fragment (Fab') directed against the CD40 ligand (CD40L, also known as CD154). Developed by Novo Nordisk, it was designed to inhibit the costimulatory signal between T cells and antigen-presenting cells (such as B cells and macrophages) by blocking the CD40L-CD40 interaction. This pathway is a critical driver of immune activation and inflammatory cytokine production in autoimmune diseases. NNC0215-0384 was primarily investigated in Phase 1 clinical trials for the treatment of active rheumatoid arthritis, specifically in patients with an inadequate response to background methotrexate treatment. The development aimed to provide a potent anti-inflammatory effect by modulating T-cell-dependent B-cell responses. However, the program is no longer listed in Novo Nordisk's active pipeline, indicating that development for this indication has been terminated or discontinued.
