NNC0385-0434 B is a specific oral tablet formulation of NNC0385-0434, a macrocyclic peptide inhibitor of proprotein convertase subtilisin/kexin type 9 (PCSK9) developed by Novo Nordisk for the treatment of hypercholesterolemia. The drug functions by binding to circulating PCSK9 and blocking its interaction with the low-density lipoprotein (LDL) receptor, thereby preventing receptor degradation and increasing the clearance of LDL cholesterol from the blood. To overcome the low oral bioavailability typical of peptides, NNC0385-0434 is co-formulated with the absorption enhancer sodium N-[8-(2-hydroxybenzoyl)amino] caprylate (SNAC). Despite achieving LDL-lowering efficacy comparable to injectable PCSK9 inhibitors in Phase 2 clinical trials, Novo Nordisk discontinued the development of NNC0385-0434 in November 2022 due to strategic portfolio considerations.
