NNC0385-0434 B is a specific oral tablet formulation of NNC0385-0434, a peptide-based biologic developed by Novo Nordisk for the treatment of hypercholesterolemia. The drug acts as an antagonist of Proprotein convertase subtilisin/kexin type 9 (PCSK9), a protein that binds to low-density lipoprotein (LDL) receptors on the surface of hepatocytes and promotes their degradation. By inhibiting PCSK9, NNC0385-0434 B prevents the degradation of these receptors, thereby increasing the liver's capacity to clear LDL cholesterol from the blood. To enable oral delivery of the peptide, it is co-formulated with salcaprozate sodium (SNAC), an absorption enhancer that facilitates transport across the gastric mucosa. Although Phase 2 clinical trials demonstrated significant dose-dependent reductions in LDL cholesterol comparable to injectable PCSK9 inhibitors, Novo Nordisk discontinued the development of NNC0385-0434 in 2019 due to strategic portfolio considerations.
