NNC0497-0040 is a long-acting amylin receptor agonist (amylin analogue) being developed by Novo Nordisk for the treatment of obesity and type 1 diabetes. Amylin is a peptide hormone co-secreted with insulin from pancreatic beta cells that plays a significant role in glucose homeostasis by inhibiting postprandial glucagon secretion, slowing gastric emptying, and promoting satiety through central nervous system signaling. As a long-acting analogue, NNC0497-0040 is designed for once-weekly subcutaneous administration, offering a more convenient dosing schedule compared to short-acting amylin mimetics. It is currently being evaluated in Phase 1 clinical trials to assess its safety, tolerability, and pharmacokinetics in healthy individuals, people with overweight or obesity, and participants with type 1 diabetes who are also overweight or obese.
