NNC5840 is a protracted glucagon-like peptide 1 (GLP-1) analogue designed to optimize cAMP-biased signaling at the GLP-1 receptor. In preclinical studies, it has demonstrated a partial-Gsα, cAMP-biased GLP-1R signaling profile in vitro, leading to superior maximal body weight reduction compared to semaglutide in diet-induced obese mice. Its weight loss profile is characterized by reduced in vivo potency but increased maximal efficacy, suggesting a novel approach to obesity pharmacotherapy.
