NNMT siRNA refers to a class of small interfering RNA (siRNA) molecules designed to silence the expression of the Nicotinamide N-Methyltransferase (NNMT) gene via RNA interference (RNAi). NNMT is a metabolic enzyme that catalyzes the transfer of a methyl group from S-adenosyl-L-methionine (SAM) to nicotinamide (NAM), producing 1-methylnicotinamide (MNA) and S-adenosyl-L-homocysteine (SAH). Upregulation of NNMT has been implicated in metabolic disorders such as obesity and insulin resistance, as well as various cancers including glioblastoma, colorectal cancer, and renal cell carcinoma, where it promotes tumor cell proliferation, invasion, and therapeutic resistance. NNMT siRNA is widely utilized as a research tool to study the biological consequences of NNMT knockdown in vitro and in preclinical animal models. As of 2024, no NNMT-targeting siRNA has advanced to clinical trials, and these reagents are primarily available through research-use-only suppliers like Santa Cruz Biotechnology and MedChemExpress.
