NNV003 is a mouse–human chimeric IgG1 monoclonal antibody targeting the tetraspanin CD37, which is highly expressed on mature B cells and many B‑cell malignancies including chronic lymphocytic leukemia (CLL) and non‑Hodgkin lymphoma (NHL).[1][4][5] It was engineered from the murine anti‑CD37 antibody lilotomab and binds the same CD37 epitope with enhanced affinity to human and murine Fcγ receptors and pH‑dependent binding to neonatal Fc receptor (FcRn), which is expected to prolong its serum half‑life relative to its murine predecessor.[1] Unconjugated NNV003 mediates anti‑tumor effects primarily through immune effector functions such as antibody‑dependent cellular cytotoxicity (ADCC) and antibody‑dependent cellular phagocytosis (ADCP), with modest direct cytotoxicity in vitro but significant survival benefits in xenograft mouse models of B‑cell malignancy.[1][5] NNV003 also serves as the targeting backbone for radioimmunoconjugates, including β‑emitter 177Lu‑NNV003 (Lu‑DOTA‑NNV003) and the targeted alpha therapy 212Pb‑NNV003, both of which deliver ionizing radiation selectively to CD37‑positive tumor cells and have shown potent anti‑tumor activity with manageable, mainly transient hematologic toxicity in preclinical CLL and NHL models.[1][4][6]
