Overview

NNZ-3006 is a synthetic macrocyclic analog of Gly-Pro-Glu (GPE), the naturally occurring N-terminal tripeptide of insulin-like growth factor-1 (IGF-1). Developed by Neuren Pharmaceuticals, NNZ-3006 was engineered to overcome the rapid proteolytic degradation and poor bioavailability associated with the endogenous GPE peptide while maintaining its neuroprotective properties. The compound is designed to modulate neuroinflammation and reduce neuronal apoptosis, primarily by mimicking the action of GPE in the central nervous system. Preclinical studies have explored its potential in treating traumatic brain injury and other neurodegenerative conditions, although it has not progressed into late-stage clinical development, with Neuren focusing its resources on other GPE-related candidates such as trofinetide (NNZ-2566) and NNZ-2591.
