No. 46 chimeric VEGF protein is a novel tumor vaccine developed through DNA shuffling of vascular endothelial growth factor (VEGF) genes from mouse, rat, human, and chicken. This chimeric protein was selected for its potent immunotherapy effects, specifically its ability to induce long-lasting and high levels of antibodies that recognize VEGF in mice. The mechanism of action involves these induced anti-VEGF antibodies blocking the tyrosine phosphorylation of the ERK1/2 pathway, which is crucial for VEGF-VEGFR interaction. This blockade leads to significant inhibition of tumor angiogenesis, slowed tumor growth, increased survival rates in tumor-bearing mice, and inhibition of lung metastases in models of H22 hepatocellular carcinoma. The drug aims to overcome immune tolerance to self-antigens like VEGF for cancer therapy.
