Overview

NO.0449-0145 is a **small molecule** inhibitor of apurinic/apyrimidinic endonuclease 1 (**APE1**), a key enzyme in the base excision repair (BER) pathway responsible for DNA repair. In preclinical studies, NO.0449-0145 inhibits APE1, leading to accumulation of DNA damage and activation of multiple cell death pathways, including apoptosis, pyroptosis, and necroptosis, in non-small cell lung cancer (NSCLC) cell lines. It is also active in overcoming cisplatin- and erlotinib-resistance in NSCLC models, and suppresses tumor progression in xenograft mouse models. This drug selectively induces DNA damage in tumor cells with high APE1 expression, sparing non-cancerous cells. NO.0449-0145 is considered a novel therapeutic candidate targeting DNA repair mechanisms for NSCLC and potentially other cancers[1][3][4][6][7].
