Overview

NO66 (also known as RIOX1 or MAPJD) is a JmjC domain-containing enzyme that functions as both a histone demethylase and a ribosomal oxygenase. It specifically demethylates histone H3 lysine 4 (H3K4) and lysine 36 (H3K36) and hydroxylates the ribosomal protein L8 (Rpl8). NO66 is frequently overexpressed in various malignancies, including colorectal cancer, non-small cell lung cancer, and osteosarcoma, where it promotes cell proliferation and the epithelial-mesenchymal transition (EMT). NO66 inhibitors are small molecule compounds designed to bind the catalytic JmjC domain, blocking its enzymatic activity to restore epigenetic homeostasis and inhibit oncogenic signaling. These agents are currently in the preclinical research stage, with various chemical scaffolds being explored for their potential as targeted cancer therapies.
