NOA-104 is a non-steroidal, multimodal small molecule drug candidate developed by Noa Therapeutics for the treatment of complex immune diseases involving barrier dysfunction, with an initial focus on atopic dermatitis. The compound is designed to simultaneously target three disease drivers—immune modulation (attenuating inflammatory cytokines), barrier restoration (upregulating epithelial and endothelial barrier proteins), and microbiome modulation (broad-spectrum antimicrobial efficacy)—to address persistent unmet needs in inflammatory barrier diseases. Mechanistically, NOA-104 acts as an agonist of the aryl hydrocarbon receptor (AHR), a transcription factor involved in regulating genetic activity related to inflammation and skin barrier function. Preclinical studies have shown that NOA-104 can modulate the immune system, restore genetic expression of barrier function genes, reduce oxidative stress in human skin cells, and resolve scratching behavior associated with itching. While its lead indication is atopic dermatitis, there is potential for expansion into other dermatological inflammatory diseases as well as systemic conditions such as ulcerative colitis, multiple sclerosis, ocular disorders, and cardiovascular indications[1][2][3][4][7].
