Nocaine is an investigational **small-molecule** piperidine-based stimulant and monoamine transporter inhibitor developed through Georgetown University Medical Center's Drug Discovery Program with Biostream and in conjunction with the National Institute on Drug Abuse. It was designed primarily as a pharmacologic substitute therapy for **cocaine addiction**, with preclinical evidence that it can substitute for cocaine in animal models while showing substantially lower abuse liability and weak reinforcing effects. Mechanistically, nocaine inhibits the **dopamine transporter** and **norepinephrine transporter**, with relative selectivity over the serotonin transporter, consistent with a DAT/NET-preferring uptake inhibitor intended to blunt cocaine withdrawal-related aversive effects.
