Nocodazole is a synthetic small molecule antineoplastic agent that acts primarily by depolymerizing microtubules. It binds to β-tubulin and disrupts the assembly and disassembly dynamics of microtubules in both interphase and mitotic cells. This leads to cell cycle arrest at the G2/M phase and ultimately induces apoptosis in susceptible tumor cells[1][2][8]. In addition to its antimicrotubule activity, nocodazole has been shown to decrease oncogenic potential via microtubule-independent mechanisms such as upregulation of LATS2 expression and inhibition of Wnt signaling[2]. It is widely used as a research tool for synchronizing cell cycles in vitro and for studying cytoskeletal dynamics[3][9]. Nocodazole is not approved for clinical use but is commonly used experimentally.
