NODX-010 is a novel, potent small molecule inhibitor of poly(ADP-ribose) glycohydrolase (PARG), developed by Nodus Oncology. It is designed to disrupt the PARylation cycle by inhibiting PARG, an enzyme involved in DNA repair processes. Preclinical studies have demonstrated that NODX-010 displays favorable pharmacokinetics and robust in vivo target engagement, leading to significant tumor regression and growth inhibition in breast cancer models. In comparative studies, it showed similar efficacy to IDE-161 (another PARG inhibitor) and outperformed the PARP inhibitor niraparib in tumor growth inhibition. The compound is currently being evaluated for its potential use in breast cancer and possibly other indications sensitive to PARG inhibition[1][3][4].
