Drug intelligence / Profile preview
The combination of nogapendekin alfa inbakicept (ANKTIVA) and pembrolizumab (Keytruda) is an investigational immunotherapy regimen designed to synergistically enhance anti-tumor immune responses. Nogapendekin alfa inbakicept is a first-in-class interleukin-15 (IL-15) receptor agonist complex consisting of an IL-15 mutant (IL-15N72D) bound to an IL-15 receptor alpha-Fc fusion protein. It mimics the natural biological process of trans-presentation to selectively activate and promote the proliferation of CD8+ cytotoxic T cells and natural killer (NK) cells without expanding immunosuppressive regulatory T cells (Tregs). Pembrolizumab is a humanized monoclonal antibody that blocks the programmed cell death protein 1 (PD-1) receptor, preventing its interaction with PD-L1 and PD-L2 to rescue T cells from exhaustion. By combining these mechanisms, nogapendekin alfa inbakicept proliferates and activates effector lymphocytes to turn "cold" tumors "hot," while pembrolizumab sustains their activity by blocking checkpoint-mediated inhibition. This combination is being developed by ImmunityBio and has been evaluated in clinical trials, such as QUILT-2.023 and QUILT-3.055, for advanced solid tumors, particularly non-small cell lung cancer (NSCLC).
