Overview

This drug combination is a chemo-immunotherapy regimen developed by ImmunityBio for the first-line treatment of advanced or metastatic non-small cell lung cancer (NSCLC). It consists of nogapendekin alfa inbakicept (NAI; Anktiva), an interleukin-15 (IL-15) receptor agonist complex, administered alongside the PD-1 checkpoint inhibitor pembrolizumab and a chemotherapy backbone of carboplatin and either paclitaxel or nab-paclitaxel. Nogapendekin alfa inbakicept is designed to enhance the innate and adaptive immune response by activating natural killer (NK) cells and CD8+ T cells through the IL-15 receptor. This "Immunotherapy 2.0" approach aims to synergize the immune-stimulating effects of IL-15 with the checkpoint blockade of pembrolizumab and the cytotoxic effects of standard-of-care chemotherapy. The regimen is currently being evaluated in the Phase 3 QUILT-2.023 clinical trial, specifically targeting patients with squamous NSCLC in Cohort B.
