ANKTIVA + Ad5 HPV is an investigational combination immunotherapy developed by ImmunityBio (formerly NantCell) for the treatment of second-line head and neck cancer, including both HPV-positive and HPV-negative cases. The regimen combines ANKTIVA (nogapendekin alfa inbakicept-pmln), an IL-15 superagonist fusion protein, with Ad5 HPV, an adenovirus-vectored vaccine. ANKTIVA consists of an IL-15 mutant (N72D) complexed with an IL-15 receptor alpha (IL-15Rα) sushi domain-Fc fusion protein; it acts as a potent agonist of the IL-15 receptor complex, stimulating the proliferation and activation of natural killer (NK) cells and CD8+ T cells without inducing the expansion of regulatory T cells. Ad5 HPV utilizes a modified adenovirus type 5 vector to deliver human papillomavirus (HPV) antigens, aiming to elicit a robust, antigen-specific T-cell response. This combination approach is designed to leverage the vaccine's ability to prime the immune system against tumor-associated antigens while the IL-15 superagonist enhances and sustains the anti-tumor effector cell response.
