Nogitecan is a **camptothecin-derived topoisomerase I inhibitor**, structurally and mechanistically related to topotecan and irinotecan. Nogitecan inhibits the enzyme **topoisomerase I**, which is essential for DNA replication. By stabilizing the DNA-topoisomerase I complex and preventing the religation of single-strand DNA breaks, nogitecan induces DNA damage, leading to cell cycle arrest and apoptosis, particularly in rapidly dividing cancer cells.
