This is a negative control antibody-drug conjugate (ADC) utilized in preclinical oncology research, specifically by Seattle Genetics (now Seagen). It consists of a non-binding humanized monoclonal antibody (isotype control) conjugated to a pyrrolobenzodiazepine (PBD) dimer, a potent DNA-alkylating cytotoxic agent. Because the antibody component does not recognize any antigen on the target cells, the conjugate serves to differentiate between antigen-specific cell killing and non-specific toxicity or systemic payload release. In studies of SGN-CD352A, this control demonstrated no significant anti-tumor activity in multiple myeloma xenograft models, confirming that the efficacy of the therapeutic ADC is dependent on CD352-mediated delivery.
