Overview

This specific formulation of aspirin is a non-enteric coated, low-dose (40 mg) tablet designed for twice-daily administration. Developed by the Montreal Heart Institute, it is currently being investigated in the COLCOT-T2D clinical trial for the primary prevention of cardiovascular events in patients with type 2 diabetes mellitus who have no known history of cardiovascular disease. The rationale for using a non-enteric coated formulation with twice-daily dosing is to address the high platelet turnover and potential 'aspirin resistance' often observed in diabetic patients, which may reduce the efficacy of standard once-daily enteric-coated aspirin regimens. As an antiplatelet agent, it works by irreversibly inhibiting cyclooxygenase-1 (COX-1), thereby blocking the synthesis of thromboxane A2 and preventing platelet aggregation and subsequent thrombus formation.
