IL-7 is a cytokine crucial for T-cell development, survival, and homeostasis, acting primarily through the IL-7 receptor composed of the IL-7Rα chain and common γ-chain (CD132)[1][3]. The non-glycosylated form maintains biologic activity, binding to IL-7Rα and activating downstream signaling to induce proliferation and survival of T cells; it preferentially expands naïve and central memory CD4⁺ and CD8⁺ T cells, thus broadening the T cell repertoire[1][3]. It has been evaluated as an immunotherapeutic agent in oncology and lymphopenic conditions, with early-phase clinical trials showing increased T cell counts and diversity, with minimal toxicity and only minor, non-neutralizing anti-drug antibody formation[2][3].
