The non-pulsed dendritic cell vaccine developed by the Hospital Clinic of Barcelona is an autologous cell-based therapy used as a control or placebo arm in clinical trials for HIV-1 infection, specifically the DCV-2 and DCV-3 studies. It consists of a patient's own monocyte-derived dendritic cells (MD-DCs) that have been extracted and matured in vitro but, unlike the active therapeutic vaccine, are not 'pulsed' or loaded with heat-inactivated autologous HIV-1 virus. In the context of these trials, this intervention serves to isolate the specific immune-stimulating effect of the HIV antigens from the general stimulatory effect of the dendritic cells themselves. The vaccine is typically administered subcutaneously to patients who are on antiretroviral therapy.
