Overview

The **non-targeted control-ADC** is a negative control agent developed by Bayer AG for use in preclinical research evaluating novel antibody-drug conjugates (ADCs). It consists of a non-binding (isotype) antibody conjugated to a pyrrole-based kinesin spindle protein (KSP) inhibitor payload via a specific linker. This control agent is designed to have the same linker and payload as targeted ADCs (such as those targeting EGFR or TWEAKR/Fn14) but lacks the ability to bind to specific tumor antigens. In preclinical assays, it serves to demonstrate the antigen-specific activity of targeted ADCs by showing significantly lower potency (often >100-fold selectivity) in antigen-positive cancer cell lines and animal models. The payload, a KSP inhibitor, targets the kinesin spindle protein (Eg5/KIF11), which is essential for the formation of a bipolar mitotic spindle, thereby inducing mitotic arrest and apoptosis in cells that internalize the conjugate through non-specific mechanisms.
