Overview
The non-targeting control amanitin antibody-drug conjugate is an experimental research tool developed by Heidelberg Pharma for use in preclinical studies, particularly those evaluating amanitin-based therapeutics like HDP-101. This conjugate utilizes a non-targeting (isotype) antibody to deliver the cytotoxic payload α-amanitin, which specifically inhibits the RNA polymerase II subunit A (POLR2A). By blocking transcription, amanitin induces apoptosis and immunogenic cell death. In the context of multiple myeloma research, this control is used to demonstrate that the anti-tumor activity of targeted ADCs is dependent on specific antigen binding (such as BCMA) rather than non-specific uptake or systemic release of the toxin.
