Overview

Non-transduced NK cells are natural killer cells that have not undergone genetic modification through viral or non-viral transduction methods. NK cells are a subset of large granular lymphocytes involved in the innate immune response, capable of killing tumor cells and virus-infected cells without prior sensitization. Their mechanism of action is primarily direct cytotoxicity via release of perforin and granzymes, induction of apoptosis through death receptor pathways (e.g., TRAIL/TRAIL-R, Fas/FasL), and antibody-dependent cellular cytotoxicity (ADCC) mediated by the CD16 (FcγRIII) receptor. These cells are regulated by a balance of activating and inhibitory surface receptors. In clinical applications, non-transduced NK cells are used in cell therapy approaches for certain cancers and viral infections, serving as a baseline for comparison to genetically modified (e.g., CAR-NK) cells[1][2][3][4].
