Nonapeptide-1 (also known as Melanostatine-5) is a synthetic biomimetic nonapeptide that acts as a selective, competitive antagonist of the melanocortin-1 receptor (MC1R). By blocking the binding of alpha-melanocyte-stimulating hormone (α-MSH) to MC1R on melanocytes, it inhibits the downstream cAMP/PKA signaling pathway, leading to the downregulation of microphthalmia-associated transcription factor (MITF) and key melanogenic enzymes such as tyrosinase. This prevents the hyperproduction of melanin and reduces skin pigmentation. Nonapeptide-1 is widely used as a skin-brightening and tone-correcting active ingredient in cosmetic formulations and is also utilized as a research tool in skin biology and pigmentation studies.
