Nonivamide, also known as pelargonic acid vanillylamide or PAVA, is a synthetic analog of capsaicin and a capsaicinoid. This small molecule is commonly manufactured synthetically and is utilized in the pharmaceutical industry for its analgesic and anti-inflammatory properties. It is primarily administered topically in creams, ointments, and patches for conditions such as muscle and joint pain, including acute low back pain. Nonivamide's main mechanism involves activating the transient receptor potential vanilloid 1 (TRPV1) receptor, leading to a sensation of heat followed by desensitization and pain relief. Additionally, it has been investigated for its role in preventing dietary-induced body fat gain and its anti-inflammatory effects, which may involve inhibiting the NF-κB/NLRP3 pathway.
