Overview

A clinical procedure involving nonmyeloablative (reduced-intensity) allogeneic hematopoietic stem cell transplantation (NST) developed and investigated at Beth Israel Deaconess Medical Center. The approach aims to treat metastatic solid tumors by using a conditioning regimen of cyclophosphamide and fludarabine to suppress the host immune system, followed by the infusion of donor-derived peripheral blood stem cells. The therapeutic effect is primarily mediated by the 'graft-versus-tumor' (GVT) effect, where donor-derived T-cells recognize and eliminate residual cancer cells. This modality is particularly explored for patients with renal cell carcinoma, breast cancer, and other solid tumors who have failed standard therapies, utilizing donor lymphocyte infusions (DLI) to further stimulate anti-tumor immunity if needed.
